In a study published in the Journal of the American Medical Association, Premarin (Conjugated Estrogens) tablets were compared to a placebo (sertraline) tablet in women with a history of atrophic vaginitis.
The study, in conjunction with an open label extension, found that pre-treatment with sertraline or placebo decreased the incidence of vulvovaginal atrophy, the most common form of vaginitis, by 16% and 22%, respectively, compared to baseline. In addition, women taking sertraline or placebo had less improvement in their sexual function than women taking either medication.
The drug, Premarin, is available over the counter at pharmacies nationwide without prescription, and has been available in many countries around the world since 1998. It is available at lower cost in both the United States and Canada, and is also available in the EU, Canada, and the European Union. The drug is also available as a topical gel over the counter at various local pharmacies.
In the study, the researchers used data from the clinical trials of the drug and its effectiveness. The participants in the study were randomly assigned to take either Premarin tablets or placebo for 12 weeks.
“The results showed that the mean number of side effects for the Premarin tablets was significantly reduced compared to those for the placebo,” said lead researcher Dr. Susan E. O’Connor, M. D., Ph. D., from the University of North Texas Health Science Center at San Antonio and Medical University in San Antonio.
“We know that there are many adverse reactions to medications in the human body that are often not well understood or reported,” she said. “This study is important because it will inform our ongoing efforts to improve treatment options for women with vulvovaginal atrophy, which is a common side effect of this drug.”
The results of the study also provide additional insight into the efficacy of this drug and how to improve its safety and effectiveness in managing vulvovaginal atrophy.
Premarin tablets contain estradiol, a hormone that helps regulate the production of folliculins and luteinizing hormone, which are naturally produced by the ovaries.
The study was supported by the National Institutes of Health and other funding sources. The authors would like to thank Jennifer K. Hargreaves, M. D., of the Division of Sexual Medicine at the University of North Texas Health Science Center and of the University of Texas Health Science Center at San Antonio for her assistance with the statistical analysis.
“This study provides the first evidence that a medication like Premarin (Conjugated Estrogens) can improve female sexual function,” said O’Connor, Ph. D., Director of the Department of Dermatology, at the University of North Texas Health Science Center.
“We’re hoping that this study will help us make better decisions about how to help women with vulvovaginal atrophy who are on medication to treat their condition.”
